Non soltanto alle persone che seguono le gesta dell'Uomo Ragno su Youtube, ma anche e soprattutto al mentore Tony Stark (Robert Downey, Jr.) che, dopo lo scontro tra storici Avengers e piccoli outsider (come Ant-Man) ha preso la questione del ragazzo a cuore. Da quando ha acquisito incredibili abilità fisiche e riflessi di ragno, Peter è passato dall'essere un adolescente ordinario, alle prese con i compiti e la scuola, gli amici invadenti, i bulli, le prime cotte, lo sguardo vigile di zia May (Marisa Tomei), a vestire i panni dell'eroe novello, lanciandosi in missioni pericolose contro avversari spietati e pieni di rancore, e all'insaputa dell'inflessibile Iron Man. Fino a quando un nemico sconosciuto di nome Avvoltoio (Michael Keaton) non minaccia la sua famiglia e i suoi affetti. Allora Spider-Man dovrà mettere da parte l'imprudenza e l'entusiasmo del ragazzino e diventare un eroe adulto e capace. Nel cast troviamo anche la cantante Zendaya, che interpreta la compagna di scuola Michelle, e Jacob Batalon che è Ned, il nerd entusiasta nonché migliore amico di Peter.
