La giovane Ayana Hirasaka alleva il mostruoso Iris - progenie, secondo antiche leggende, dei demoni dell'inferno - per vendicarsi, a tempo debito, di Gamera che ritiene responsabile della morte dei suoi genitori. Quando le forze armate decidono di sbarazzarsi di Gamera il cui spirito battagliero si volge adesso contro i mostruosi Gyaos scatenando devastazioni nel paese, Ayana guida telepaticamente Iris contro di lui. Tokyo, ancora una volta, farà da scenario ad un epico duello nel quale la posta in gioco è la sopravvivenza stessa dei giapponesi.
La storia corre sui binari della tradizione dei film sui mostri, sviluppando come di consueto complesse sottotrame e suggerendo simbolici richiami alla salvaguardia della natura e al valore dei sentimenti. La novità è rappresentata dall'accuratezza degli effetti speciali che, secondo alcuni, rivaleggiano con quelli del Godzilla di Emmerich: non più mostri di cartapesta, goffi e puerili, ma creature generate con la computer graphic e terremoti e distruzioni di grande spettacolarità. Il film perde l'ingenua poesia del prodotto artigianale, ma guadagna consensi presso le nuove e più esigenti generazioni di spettatori.
